Are you asking of the specific mark of the beast or just any general "mark of evil"? As for 666, what we know of it is that it is directly tied to the identity of the beast in some way, but ultimately, it is a mystery to us in how the connection works. One interpretation I know of about it is that, just as the number seven is used to mark completion in many stories of scripture (seven days of creation, seven pairs of the clean animals on the ark of Noah, seven bowls of the wrath of God poured onto our planet in the Great Tribulation), a trinity of sixes, the number of man, who was created on the sixth day (therefore a trinity of these numbers being the vile man's effort to impersonate God, who operates through His Trinity), is a representation to Satan's ultimate failure in His claims on God's power. Besides that, it could be interpreted in many other ways. The hexagram used on the flag of the State of Israel, for example, has six sides within it, six triangles upon it, and six points at its ends. There's a lot of ways to see it, but we aren't able to comprehend this mystery for certain. We know it is tied to evil, of course, but we won't know how exactly it functions as the number of the beast until the Great Tribulation itself.
